Vitrakvi is designed as a kinase inhibitor for adult and pediatric patients with solid tumors that:

• have a NTRK gene fusion without any known acquired resistance mutation,

Recommended Dosage: The recommended dosage of Vitrakvi capsules for adult and pediatric patients with body surface area (BSA) of minimum 1.0 meter-squared is 100 mg and pediatric patients with BSA not more than 1.0 meter squared is 100 mg/m2 orally twice daily, with or without food, until disease progression or until unacceptable toxicity.  

Swallow capsules of Vitrakvi whole with water. It is not advisable to chew or crush the capsules. Avoid making up a missed dose within six hours of the next scheduled dose. If puking occurs just after taking a dose, take the very next dose at the scheduled time.

• Central nervous system (CNS) side effects may occur in patients receiving Larotrectinib, including mood disorders, dizziness, cognitive impairment, and sleep disturbances. Patients should not drive or operate machinery in case of neurologic side effects.
• Quickly assess patients with signs or symptoms of fracture (e.g., changes in mobility, deformity, pain). There are no findings on the effects of Larotrectinib on healing of known fractures or risk of future fractures. 
• Those who received Vitrakvi 100 mg, increased AST of any grade may occur and increased ALT of any grade may occur. Assess liver tests, including AST & ALT, every 14 days during the 1st month of therapy, then monthly thereafter. 
• On behalf of findings and  mechanism of action, Larotrectinib can cause fetal harm if used by a pregnant woman. Women of reproductive age should use a relevant method of contraception during therapy and for 7 days after the last dose of Vitrakvi.
